So, this topic lends itself well to scripted single player events. But I think some of the best video game moments come in multiplayer.
Like anyone who is a a PC gamer, I played a lot of Counter-Strike (back in high school). I can still remember some epic rounds where I single-handedly killed 8-9 players on the other team. The nature of the game, where everyone dead is watching you dominate, makes it a pretty awesome when you pull it off.

Not really an epic moment in the game, but in Red Dead Redemption when you cross into Mexico for the first time and Jose Gonzalez' "Far Away" starts up.

I just started that game over and when I landed in Mexico I whistled for my better horse from America instead of the one the game gives you and the song didn't play.
Easily one of my favorite games. I remember when I first loaded it up and you are in the tram and I am patiently watching everything go by and by accident I bumped the mouse and noticed I could move around. It sounds stupid, but I was shocked. Every other game I had played up until that point wasn't like that. It was usually a cut scene, training, then normal game. Half-Life put all those things right into the game play. Such a great experience that game was.
The first (modern) game I every really played on a computer was Myst on my uncles Mac. Right at the moment I solved the first puzzle - the stars in the planetarium thing was glorious.
That afternoon was amazing and got me hooked on computers.

taking my first head with the demoman's "Eyelander" sword in TF2, and hearing him scream "There can be only one!"
Getting my first taunt kill in TF2 was pretty memorable.
There was one especially memorable point in Half-Life 2 for me. After battling in Ravenholm (for a few gaming sessions, I got stuck), finally emerging from the dark back into the sunlight. Something about the lighting, music and general atmosphere really impressed me.

When I finally beat Battletoads for NES. What a freaking hard game.
Dude, you're my hero. Battletoads and Ninja Gaiden were probably the two hardest games I've ever played on the NES. I could never get past the second speed bike stage on Battletoads.
