Just go and talk to your boss. He can't expect you to be his admin assistant forever. Plus, if he's a normal guy, he'd be happy that you are looking to move up.

He's a great guy so I'm sure he'll understand. His term is only for 4 years so after that someone else will replace him.

Yeah I agree with Frank. I'm not completely sure about this, but I would think it would be easier to find someone to come in and fill your position as an assistant, than find someone right for the HR Manager position.
They want someone right for the higher-up position and promotions from within are some times the best route to go. I don't think your Director is going to have any negative opinions about you wanting to do better for yourself in the company.
