I think it's really difficult for anyone to be receptive to any strangers for any purpose on the train. It's just not where your 'train' (pardon my pun) of thought is at the time. Public transit also has this 'prolitariat' feel to it that somehow makes it seem even dirtier to explain to your friends that you met someone on the train.
As for picking up women on the train, the only time I've ever tried was when some drunk homeless guy was wrecking havoc for the general populace on the train after normal working hours (I think it was like 8 or so at night). The homeless guy approached this girl and started trying to talk to her in his broken speech. She looked thouroughly disguisted with him, so I started up a convo with the homeless guy to distract him until his stop at the end of the free fare zone downtown. Afterwards it seemed to be a nice lead in to a conversation with her, got a phone number! Seems to me it was made easy only due in part by a window of opportunity to talk to her without looking like some guy trolling for his next prey.
Hmmm this could become a team sport. Have a buddy play the homeless guy and the world could be yours!
The trick for picking up in public is INVESTMENT. Don't think of the pickup as a single day event, it is a long term deal. This is how it works at the office, office tower, C-Train etc. Once you recognize that the girl you are into is in your office or takes your bus, you simply start with a hello, and don't press it. After a couple of hellos, you simply ask what they are all about (do they work here, etc). You have basically broken the ice simply by being polite. You then are not strangers, so that when you finally get the chance to chat it is much easier to gauge their attraction or interest.
Once this is mastered, picking up in assorted locations is easy.
This method certainly does not work on a single encounter, but if you practice what I preach, you should be well enough trained to advance to the next level.
So far my method of pulling the safety cord when they're running and then politely asking if they'd like some first aid after they've flown off the treadmill isn't working so well.

So basically you need to do a lot of hardcore stalking (same bus, same location, same time, again and again) to appear more natural and less creepy in the eyes of the girl, than if you hit on a her on the spot just because you thought she was hot.

I sat next to a really cute girl this morning on the bus. But I was half dozing in and out, so I probably looked like quite the weirdo (especially in my cowboy duds trying to keep my head up and constantly drifting to one side). We did the whole 'look exchange' thing, and then concidentally she followed me to work... turns out she works in the Shell building, which is right next to mine!
Maybe on Monday I'll follow her to work. I'll try and grow a mustache in the meantime. And bring a camera. Hopefully that seals the deal.

I find that mustaches and cameras are always the best way to pick up women. Also, jumping out of bushes. Lots of jumping, lots of bushes. Always with a camera and a mustache.

No no... I don't think lots of bushes really work... small bush is a better bet.
I've had great success by making eye-contact and then giving a guilty shy smile followed immediately by snapping my fingers and giving her my patented "DOUBLE GUNS" while saying "heeeeeeeeeeey"

...I forgot to add that after I give her the "DOUBLE GUNS" I open the side door of Sex-a-Tron (the name of my van) and invite her in.
I happen to know Two Fivenagame personally and can vouch for this approach working. I can also vouch for Two Fivenagame's ability to pick up a girl by doing nothing more than letting her cut his hair.
