Had a VP at a place I used to work who if you didn't lock your computer and left for lunch hour. He'd go into your Outlook, send an email to guys and gals about how you have a secret crush on them. You wouldn't know the email was sent, and go talk to one of the people after the email was received and their acting all weird and trying to have a conversation with you to let you know they like you but not that way.
God I hate practicle jokes such a waste of time. If people only put half the effort into their job as they do their jokes.

For our department it's usually a gay unicorn desktop picture. Although one time one of the guys sent out an email inviting a bunch of co-workers over for a Dungeons & Dragons party. That one was rather amusing.

Back in the day I had changed the admin girl's desktop background to that of Star Wars Kid. She eventually figured out how to change it but for some weird reason, everytime she booted up her computer, Star Wars Kid flashed on her screen for 2 seconds until her normal background came up. That was awesome.
Another prank I used to do was I'd leave some random technical textbook on the new guys desk with a note from my boss to read some random chapter or journal article. I'd make sure my boss had left for the weekend so that they couldn't clarify it with him either. Fun times.

Something simple for those slightly computer illiterate people you work with, I did this last year. Guy leaves his desk so I sat down and grabbed a screen shot of his desktop, hid all the icons and the task bar, replaced his wallpaper with the screenshot. Laughed from the down the hall while he cursed at his computer for the next 15 minutes.
Once I also took all the keys off another guys keyboard and put them in alphabetical order instead. Which only works for people who need to look at their keyboard to type since all the keys still do the same thing.

Another easy fun one is Ctrl + Alt + down arrow. It's fun watching people panic and have to call IT.

Back in the day when the internet consisted of logging into a unix account, my friend had given me his username and password to help clear out some junk from his user account.
You used to be able to set the login script to display a message, so I decided to write a pretty formal looking one that basically said he was being expelled from university immediately for disclosing his login credentials.
He freaked out, frantically asking me if I gave anybody his password and if I knew what this message meant. Of course I said no, I had no idea. So he calls the university computer department to beg not to be expelled for this.

Swapping all the keys is much too obvious for the hunt-and-peck typist. You just switch one or maybe two sets of keys, like swap m and n and maybe s and d.
Thought of a good joke: you can add custom words to people's AutoCorrect dictionary in word. Your imagination is the limit on how far you take this one (and how badly you screw someone).

I've got a lot of good mileage out of this one the last few years. Sent it to my dad at work 3 years ago, and he just called me and asked "What the F is wrong with you?" and hung up.
The best reactions are when you see the guy panicking trying to figure out how to turn it down. It used to be on youtube, but I can't seem to find it anymore. It definitely needs an more modern pop culture reference as an update.

I guy I used to work with fell for this one. Five of us worked in the same giant sized office and he was trying to listen to the clip, cranked the volume to the max and then just as he finished asking if anybody else can get try to get this video to work... well, the poor guy just about had a heart attack and the rest of us nearly died from laughing so hard.
