Mobile is cool but a rather rough city, if you're a big football fan you can take a trip up to Tuscaloosa and then Moundville Alabama for the Native American stuff. Horseshoe bend is by Montgomery too (war of 1812).

My wife and I drove from New Orleans to Pensacola Florida over a few days a couple of years ago. The best part was Gulfport/Biloxi Mississippi.
If you want to gamble, there's plenty of casinos there. Good southern food too.

Not much to look at from the outside, but lots of 'colorful' locals inside, and friend food out the wazoo. Definitely an experience.

Don't go to Jackson. Scary city. Empty streets downtown at 4:30 in the afternoon. Got honked at by the (rare) other cars. Probably not a good sign.
My top recommendation in Mississippi is Oxford. Unfortunately it's a whole lot closer to Memphis than New Orleans. Great town, home of Ole Miss.
As for the gulf, Biloxi (pronounced Buh-lux-ee btw, which was a surprise to me) is good if you're into gambling. Mobile was nice, reminded me some of Victoria actually. Best part of the gulf though (outside of nola), IMO, is Pensacola. Fun town, nice beaches, cheap everything.
As for little towns, I enjoyed Bay St. Louis, MS. It was devastated by Katrina, but the town has come back. You can really tell there's a sense of community pride. Nothing special to see, but I thought it was a fun small town. We went to a nice restaurant on the gulf right by the bridge to Pass Christian (I think it was called trapani's).

Go to Natchez for some donuts then head north on 61 to Lorman for some fried chicken at the Old Country Store. It was featured on Alton Brown's Feasting on Asphalt a few years ago and after trying it this summer, I can't say I disagree with him about it being the best fried chicken in the world (though Willie Mae's Scotch House in New Orleans might get the nod if you like your chicken to have a little kick).
From there, continue north to Rosedale, MS for some tamales at the White Front Cafe (surprisingly, the area is known for them) and stop for the night in Memphis. Interstate Barbecue is famous and certainly worth a visit, but you absolutely need to get a pork sandwich at Payne's.
The next day, you can loop southward through Oxford, Tuscaloosa and/or Biloxi on your way back to New Orleans.

All spice and no flavor, plus the original dreamland is in a terrible area and the shack is nasty. Nasty as in how in the hell did it pass a health inspection.
Archibalds is much better along with Jim n' Nicks. Chucks Fish in Tuscaloosa is a fantastic restaurant.
BTW if you come down here and make it to Tuscaloosa hit me up I will show you around town.

I was only ever in Huntsville, but that was a pretty neat little town. I'm hoping to do a trip to Tuscaloosa next year for a Bama game.
