What type of wood? If it's pine or something soft it will have penetrated probably more deeply, but enough sanding will take it off. I'd start at 120 grit and work finer after you have removed it.
If you want darker stain you need more coats, not heavier coats. If it is soft wood make sure you use a wood conditioner first, it will prevent blotching.
