When Gore lost Florida, it was the state supreme court that nixed the recount. There was some talk at the time that he could take it to a higher level, but he didn't because the process would be too damaging and had a low chance of success anyway.

Yes it can, if it's for Constitutional issues. The Supremacy Clause itself comes from Article III of the Constitution. Three amendments directly involve voting rights. The Supreme Court has ruled on these issues before (many times). Plus, there are many federal laws that describe voting accessibility.
I think the real question is to what extend can the process be expedited.
