The big ones are ok, if you haven't seen them. I prefer the smaller ones. Last year we looked on kijiji and there is a service you can pay $40/hr and have a guy take you in your vehicle to any area you want at your pace. Way better than a bus full of strangers. All you need is one moron to ruin your experience.
I have seen a lot of the wineries in that area but my favourite area is Lake Country near Winfield. Grey Monk and many others, IMO, are way better than Mission Hill. Way less pompous. I always did like Quails Gate though.
