They were trying to work out who lived in a big house in Attabad, they figured if they could get DNA from the kids in the house they could tell if it was Osama Bin Laden, to do this they set up a fake vaccination program in the town, went around offering vaccinations in some neighbourhoods, putting up posters etc order that when they knocked on the door of Osamas house they could get DNA from the five or six kids in there and see if they were bin ladens. It didn't work as the kids mothers wouldn't allow them to get vaccinated anyway.
Other than the subsequent backlash the whole idea was an elegant way to avoid just blowing a house to hell with no knowledge of who was in it, which was the alternative being proposed. By CIA standards it was incredibly light footed ironically.
