That's not necessarily true. When Lex was first introduced, he was very much the older mad scientist with no connection to Superman. When the Silver Age came along, Lex was reintroduced as knowing Superboy as he grew up in Smallville. He blamed Superboy for a fire which caused him to go bald, and that's how their rivalry grew.

When the DC universe was rebooted following Crisis on Infinite Earths in the late 80's, Calgary born writer John Byrne created a new origin for the Man of Steel, coincidently in a title named the same. In this, Luthor had no connections to Smallville, but was an older, successful businessman and scientist.

In 2003, Mark Waid wrote "Birthright", which helped reboot Superman's origins as there had been continuity altering events in the DCU. In this storyline, this is where Luthor famously became engraved in the Superman lore, as he and Clark Kent were childhood friends. He had no knowledge of Clark's Kryptonian powers, and Superboy never existed. As Clark grew older, only he knew the connection between Luthor and Smallville.

Lex's connection to Superman's past has changed significantly throughout the years.
