The Wynn is a great hotel, as is the Venetian.
The Venetian is a little more central, but only by half a block or so.
The Venetian has a little more variety in terms of shopping and restaurants.
The Wynn is very high scale and we found there aren't really any places to grab a meal under $20 a person, even if you want something quick/lite. (the Ventian has some cafes that have reasonable rates)
The Wynn has an older crowd, as mentioned, and it has higher minimums at most of their tables, but I found their waitresses to give better service as long as you tip a buck a drink.
The drinks around the pool at the Wynn are $14+ so beware.
