When Hugh Laurie came in to do his audition, he came in already "in character" and when he was done, the director commented on how perfect he was.
I would have loved to see their faces afterwards when he started talking to them in his normal British accent.

Hugh didn't walk in for an audition, nor did he start in character. He was in in Namibia while filming Flight of the Phoenix in the desert. He videotaped himself in the bathroom because he couldn't get proper light anywhere else. He also started with his British accent apologizing for the bathroom and how he looked (dischevelled since he was playing a guy marooned in the desert)...which turns out to be the House look....
He then got into the House character (which he thought was a sidekick character and that Wilson was the lead character since he was the "young-dashing" Doctor). He never expected to be auditioning for a lead role. He mailed the videotape to the U.S. where Bryan Singer leapt to his TV screen seeing the audition tape thinking that House was an American.
