As someone who hated the ending, ME3 is still an amazing game. The entire series ranks at top of my list. I feel it's a must play. As a piece of science fiction, Mass Effect has the deepest and most fleshed out universe since Star Wars.
Bioshock is a fantastic game as well and another I'd suggest is a must play. However, you won't get more than 20 hours playing through Bioshock 1. I haven't played 2 yet but I doubt there is more than 15 hours to that game either.
Dishonored is great too. I've logged somewhere around 40-50 hours with the base game plus all of the DLC. That essentially includes two separate playthroughs. It's a lot like Bioshock with maybe a little Assassin's Creed thrown in.
I'd probably rank them thusly,
Mass Effect
Bioshock
Dishonored
Red Dead Redemption (I'd expect this to jump up a spot or 2 by the time I've finished it)
Hitman
Fallout 3
