I don't see why not but you could just keep the zipped or unzipped game roms stored in a folder on your PC if you have the space in case you needed to create a new microSD someday.
Now that I think about it more you'll use a usb thumb drive to transfer the roms to the Pi. There are other ways, but this is what I did

Not much point in having a backup of Retropie as you'd probably want the latest update anyways
